Mass spectrometry (MS) solves the most critical hurdles in antibody characterisation by providing a high-resolution map of structural heterogeneity that traditional assays miss. It precisely sequences an antibody and localises complex post-translational modifications (PTMs) like glycosylation and oxidation, which are vital for ensuring drug safety, efficacy and potency.
Integrating Intact Mass, Top/Middle-down and Bottom-up MS provides a multi-level solution to the uncertainties in biophysical properties of antibodies. Intact mass analysis offers a global "bird's-eye view," rapidly identifying major glycoforms and confirming the overall molecular weight to detect and measure large-scale degradation, truncation and impurities. Bottom-up MS at the peptide level provides high-sensitivity sequence confirmation and precise localisation of PTMs like oxidation or deamidation. Bridging the two, Top/Middle-down MS interrogates intact proteins or large subunits in the gas phase, preserving relationships between modifications and proteoforms and revealing how multiple PTMs coexist on a single molecule—a feat not possible with peptide-level analysis alone.
